为进一步提高武夷菌素的产量,以基因工程菌Streptomyces ahygroscopicus var. wuyiensis W273为试验菌株,采用单因子试验确定发酵培养基的最适6种营养成分为玉米粉、葡萄糖、黄豆面、氯化铵、碳酸钙和氯化镁;应用SAS软件中的Plackett-Burman设计法,对影响菌株发酵生产武夷菌素的6个因素进行了筛选,确定葡萄糖和碳酸钙为影响摇瓶发酵生产武夷菌素的主要因素。利用响应面分析法对2个主要因素的最佳水平及其交互作用进行研究,建立了二次回归方程,Y=218.18+270.28X1+1274.74X2-3.28X12-5.4X1X2-200.98X22,并最终确定最佳培养基配方为玉米粉20 g/L、葡萄糖39 g/L、黄豆面20 g/L、氯化铵4 g/L、碳酸钙2.65 g/L、氯化镁0.4 g/L。以新配方在28℃、220 r/min进行摇瓶发酵60 h后,武夷菌素效价平均值为7215 μg/mL,较原始培养基发酵后武夷菌素效价提高了26.5%。 相似文献

The objective of the present study was to investigate the effect of fermented apple diet (FAD) supplementation on the growth performance and meat quality in finishing Berkshires. The FAD was made from dropped apple mixed with rice bran and barley bran. Until 81 ± 1 kg live weight at 133 ± 1 days, the animals were fed a growing diet, after which experimental samples were fixed at 0, 2, 4 and 6% FAD as C, T1, T2 and T3 in the finishing diets. Growth performance, ADG, ADFI and feed efficiency were improved in T1 than other groups. In carcass parameters, carcass weight was higher ( P < 0.05) in T1 than in other groups. In meat quality, moisture and crude protein contents decreased ( P < 0.05) by addition of FAD. pH24 and WHC were higher ( P < 0.05) in T1 than other groups. In sensory evaluation, marbling of fresh meat and tenderness, juiciness, flavor and overall acceptability of cooked meat were improved by the addition of FAD. The purpose of this study was to describe relevant canine brain structures as seen on T2-weighted images following magnetic resonance (MR) imaging at 7 T and to compare the results with imaging at 1.5 T. Imaging was performed on five healthy laboratory beagle dogs using 1.5 and 7 T clinical scanners. At 1.5 T, spin echo images were acquired, while gradient echo images were acquired at 3 T. Image quality and conspicuity of anatomic structures were evaluated qualitatively by direct comparison of the images obtained from the two different magnetic fields. The signal-to-nose ratio (SNR) and contrast-to-noise ratio (CNR) were calculated and compared between 1.5 and 7 T. The T2-weighted images at 7 T provided good spatial and contrast resolution for the identification of clinically relevant brain anatomy; these images provided better delineation and conspicuity of the brain stem and cerebellar structures, which were difficult to unequivocally identify at 1.5 T. However, frontal and parietal lobe and the trigeminal nerve were difficult to identify at 7 T due to susceptibility artifact. The SNR and CNR of the images at 7 T were significantly increased up to 318% and 715% compared with the 1.5 T images. If some disadvantages of 7 T imaging, such as susceptibility artifacts, technical difficulties, and high cost, can be improved, 7 T clinical MR imaging could provide a good experimental and diagnostic tool for the evaluation of canine brain disorders. 相似文献

The effects of migrating seniors on the provision of local public services in rural communities is growing in importance because of the large number of retiring baby boomers and the increasing rate at which these retirees are locating outside traditional retirement destinations. Some communities are optimistic about attracting and retaining retirees as an economic development strategy, but others are concerned that inmigrating seniors may be reluctant to support local public services, such as education, bringing with them “Gray Peril.” This article attempts to clarify questions regarding the Gray Peril hypothesis and local ability and willingness to fund education in Tennessee, an increasingly popular retirement destination. To this end, county per pupil education expenditure growth is explained by growth trends in local property tax assessment and sales tax revenue, and migration patterns of the retirement‐aged population from 1962 to 2002. 相似文献

An effective adsorbent for the removal of heavy metals was manufactured by immobilization of jujube powder. The adsorptions
of Cd, Zn and Cu from aqueous solutions by jujube complex beads (Type 1 and Type 2) were studied in a batch adsorption system.
The adsorption data were fitted well with the Langmuir isotherm models. The adsorption capacities (β) for Cd, Zn and Cu were 4.23, 2.93 and 3.64 mg/g in Type 1 and 1.24, 0.70 and 1.35 mg/g in Type 2 beads. The removal efficiencies
of the Type 2 beads, with a larger unit surface area, were lower than those of the Type 1 due to part of the casein or cyclic
AMP being destroyed during the drying process of the Type 1. These values for Type 1 beads were higher than those of all other
adsorbents for each heavy metal. A comparison of the kinetic models on the overall adsorption rate showed that the adsorption
system was best described by pseudo-first-order kinetics. The removal efficiencies of Cd, Zn and Cu exhibited similar tendencies
to those observed in the equilibrium tests. This indicates that the jujube complex beads developed in this study can be used

In the present study, the applications of mesoporous materials based on silica, namely post-synthesized, one-pot synthesized, and pure MCM-41, were investigated for the removal of phosphate from aqueous solution. The mesostructures were confirmed by X-ray diffraction, Brunauer?CEmmett?CTeller, Fourier transform spectroscopy, and transmission electron microscopy. The absorptions of phosphate by the mesoporous adsorbents were examined, with different adsorption models used to describe the equilibrium and kinetic data. The maximum adsorption capacities of the mesostructure adsorbents were found to be 45.162, 40.806, and 31.123 mg g?C1 for the post-synthesized, one-pot synthesized, and pure MCM-41, respectively. The kinetic data showed that the adsorptions of phosphate onto the post-synthesized and pure MCM-41 followed the pseudo-first-order kinetic model, whereas the one-pot synthesized adsorbent was described by the pseudo-second-order model. 相似文献

A pot experiment was conducted in a plastic film house to evaluate the translocation and uptake of heavy metals(Pb,Cd,Cu,and Zn) into brown rice(Oryza sativa L.) and the heavy metals residues in soils which had previously been irrigated with domestic wastewater for a long time(3 years).The range of Pb,Cd,Cu,and Zn was 5.10 ± 0.01,0.105 ± 0.017,5.76 ± 0.42,and 23.56 ± 1.40 mg kg-1,respectively in the domestic wastewater-irrigated soil,and 0.370 ± 0.006,0.011 ± 0.001,0.340 ± 0.04,and 2.05 ± 0.18 mg kg-1,respectively,in the domestic wastewater-irrigated brown rice.The results indicated that application of domestic wastewater to arable land slightly increased the levels of Pb,Cd,Cu,and Zn in soil and brown rice(P < 0.01).The concentrations of heavy metals in brown rice were lower than the recommended tolerable levels proposed by the Joint FAO/WHO Expert Committee on Food Additives.However,the continuous monitoring and pollution control of hazardous materials from domestic wastewater are needed in order to prevent excessive build-up of heavy metals in the food chain. 相似文献
